セリン欠乏はin vitroではESCC細胞の増殖を効果的に阻害したが、セリン欠乏（-Ser）食またはセリン/グリシン欠乏（-SG）食単独ではin vivoでの腫瘍の成長を阻害しなかった。この結果は、-SG食がHCT116、DLD-1 [ 37 ]、MC38 [ 48 ]、自然発症膵臓癌モデル [ 26 ]、および原発性乳癌細胞 [ 49 ] を含むさまざまなin vivoモデルで腫瘍の成長に影響を与えなかったことを示す以前の研究と同様である。SSPの活性化がこの結果の主な理由であると思われる。これらの結果は、セリン飢餓がATF4を介したSSP活性化を有意に促進するという我々の観察と一致した。このように、我々の結果ではこの組み合わせがin vivoでESCC腫瘍の成長を有意に阻害することを示したため、セリン欠乏とPSAT1ノックダウンの組み合わせはESCC治療に効果的な戦略である。
要約すると、本研究は、セリンがDNA合成と酸化還元恒常性を維持することでESCC細胞の増殖を促進することを実証し、セリンがmTORC1の活性化を維持する新たなメカニズムを明らかにした（図7J ）。これらの知見は、ESCCの潜在的な治療戦略として、セリン代謝を標的とする阻害剤の開発に向けた信頼できる理論的根拠を提供する。
